At least 10 of the people hurt in the accident were rushed to a nearby hospital.
A multi-agency response involving police, fire, ambulance and Network Rail staff took place, Glasgow Live reports.
The bus belonged to national bus company First Bus.
A spokesman for Scottish Fire and Rescue said: “We were alerted at 11.39am on Sunday the 21st May to reports of a road traffic collision involving a bus in Glasgow.
"Operations control mobilised three appliances to Cook Street where firefighters were supporting their partners to make the area safe.
"Three casualties have been transported to the Queen Elizabeth Hospital by the Scottish Ambulance Service.
"A number of additional casualties have been treated at the scene but were not required to be taken to hospital.”
The area was closed off, with Police Scotland advising the public to avoid it and “use alternative routes”.

The route was also closed for trains going between Glasgow Central and Paisley Gilmour Street while the damage to the bridge was assessed, however it was reopened at around 2.40pm.
A spokesman for First Bus said: "We can confirm that one of our buses was involved in the incident on Cook St, earlier today.
"We are working closely with the emergency services at the scene and our thoughts are with those involved in this accident.”
It is not clear how the accident happened, given there are four warning signs about height restrictions around the bridge.
No update has been given on the condition of those who were taken to hospital.
